On-line commissioning of SHIPTRAP

Abstract The on-line commissioning of the Penning-trap mass spectrometer SHIPTRAP was successfully completed with a mass measurement of holmium and erbium radionuclides produced at SHIP. A large fraction of contaminant ions created in the stopping cell was identified to originate from the buffer-gas supply system. Using a liquid nitrogen cold trap they were reduced to a tolerable amount and mass measurements of Er 147 , Er 148 , and Ho 147 with relative uncertainties of about 1 × 1 0 − 6 were performed.

Droplet microfluidics is a technology that enables the production and manipulation of small volumes. In biosciences, the most popular application of this technology is Droplet Digital™ PCR (ddPCR™), where parallel nanoliter-scale PCR assays are used to provide a high sensitivity and specificity for DNA detection. However, the recovery of PCR products for downstream applications such as sequencing can be challenging due to the droplets' stability. Here we compared five methods for disrupting the droplets to recover DNA. The Use of 15N NMR for the Understanding of Nitrogen Physisorption.

Abstract Nitrogen adsorption at liquid nitrogen temperature, 77K, is universally employed to determine pore dimensions. The nature of nitrogen physisorbed within the micropores of ZSM-5 zeolite was studied by in situ15N NMR as it depends on the partial pressure of the nitrogen and the temperature above and below 77K. An increase in the volume of adsorption of nitrogen at 77K occurs at a relative pressure of ~0.18 for this zeolite. The nature of the nitrogen was studied below and above this increase in nitrogen adsorption. We find that two states (phases) of nitrogen are evident. 3-D imaging and quantification of graupel porosity by synchrotron-based micro-tomography

The air bubble structure is an important parameter to determine the radiation properties of graupel and hailstones. For 3-D imaging of this structure at micron resolution, a cryo-stage was developed. This stage was used at the tomography beamline of the Swiss Light Source (SLS) synchrotron facility. The cryo-stage setup provides for the first time 3-D-data on the individual pore morphology of ice particles down to infrared wavelength resolution. A rapid and cost-efficient DMSO-based method for isolating DNA from cultured lichen photobionts

We have developed a simple and fast procedure for the purification of PCR-quality DNA from cultured lichen photobionts. This new one-step method uses the solvent dimethyl sulfoxide (DMSO) combined with heat treatment to chemically breakdown algal and plant tissues. The DMSO-DNA extracts may be directly precipitated and purified by standard techniques in a total time of approximately 30 min. Compared to other DNA extraction protocols, the DMSO-based method suppresses the need for liquid nitrogen, any extraction buffer, grinding, phenol, and long incubation or centrifugation steps, thereby considerably reducing the possibility of contamination. Comparing encapsulation-dehydration and droplet-vitrification for cryopreservation of sugarcane (Saccharum spp.) shoot tips

Abstract In this study, in vitro shoot tips of two sugarcane clones were successfully cryopreserved using encapsulation-dehydration and droplet-vitrification with two vitrification solutions, PVS2 and PVS3. For both clones, encapsulation-dehydration induced significantly higher recovery, reaching 60% for clone H70-144 and 53% for clone CP68-1026, compared with droplet-vitrification in which recovery was 33–37% for clone H70-144 and 20–27% for clone CP68-1026. Optimal conditions included preculture of encapsulated shoot apices for 24 h in liquid medium with 0.75 M sucrose and dehydration with silica gel to 20% moisture content (fresh weight basis) before direct immersion in liquid nitrogen. …

Charge Mobility and Dynamics in Spin-Crossover Nanoparticles Studied by Time-Resolved Microwave Conductivity

We use the electrode-less time-resolved microwave conductivity (TRMC) technique to characterize spin-crossover (SCO) nanoparticles. We show that TRMC is a simple and accurate mean for simultaneously as-sessing the magnetic state of SCO compounds and charge transport information on the nanometre length scale. In the low-spin state from liquid nitrogen temperature up to 360 K the TRMC measurements present two well-defined regimes in the mobility and in the half-life times, possessing similar transition tempera-tures TR near 225 K.
